Verification and/or validation of test methods are essential aspects of a laboratory’s work in accordance with UNE-EN ISO/IEC 17025.

To carry out validations, there are different techniques, and we highlight the 3 fundamental ones:

  • Comparison against a stable and known value using a reference material available in sufficient quantity for quantification.
  • Laboratory preparation of the known reference value using standards and doping a matrix of interest.
  • “Validation by pair values” is employed when we cannot resort to the other two, either due to the impossibility of doping the samples or the lack of quantity to perform sufficient repetitions with reference materials.
Laboratories should perform validations following a documented procedure and use the above methodologies as appropriate. To implement any of the techniques, calculations need to be performed to confirm that established prerequisites are achieved that determine whether the validation is fit for purpose or not. By checking certain validation parameters such as precision (in repeatability and/or reproducibility) and accuracy, as the most prominent of the validation. In addition, another important requirement when performing validation calculations for a test method when working according to UNE-EN ISO/IEC 17025 is the determination of the test uncertainty. There are different methodologies for its calculation and some of them can be complicated. For this reason, it is recommended that the laboratory has a procedure and tools that allow its evaluation to be carried out in a simpler way and minimising the possibility of calculation errors. The results of the experiments performed during validation allow, by comparison with the prerequisites, the declaration of the validation of the test method and the establishment of acceptable values for quality controls.
